Thank you for posting.
In terms of meaning, both will be equally understood. However, as used in the dialogue [Que horas preciso deixar o quarto?] is more used in daily conversations, casually.
However, in writing, it would be good to use the preposition, just as it is used in the answer for this type of question, for example:
[A que horas comeรงa a aula?]
[(At) What time does the class start?]
[A aula comeรงa ร s 8 horas.]
[The class starts at 8 o'clock.]

Is there any difference between saying "A que horas preciso deixar o quarto?" and "Que horas preciso deixar o quarto?"
